The phrase "... her seed" is referring to offspring from Eve. As woman do not have seed, Genesis 3:15 gives us a hint of the special and unique birth of the coming one that would "bruise the head" of Satan, defeating sin and death and providing a way of atonement for all that accept Him. Reference Isaiah 7:14a, "The Messiah would be born of a virgin."

Near the end of his life, Jacob assembled his twelve sons and blessed them and prophesied about the future of each of them. In Genesis 49:8-12, Jacob, speaking to Judah, says that rulership will come through his descendants. Thus the primary identifiers of the Messianic line were Abraham, Isaac, Jacob, Judah, and then David. Jesus, the ultimate objective and culmination of the Messianic hope, was in this same genealogical line. In Genesis 49:9 Jacob compares Judah to a lion, and in Revelation 5:5 Jesus is called "the Lion of the tribe of Judah."

Shiloh means "the one who brings", or "until he comes to which it belongs" and is a word commonly attributed as a name for the Messiah. Ezekiel 21:27 uses the same phrase when addressing Zedekiah (the last king of Judah) stating that Israel would not have a king again until Messiah came. The "scepter" represents the authority and right to rule (see Numbers 24:17,  Esther 4:11,  Isaiah 14:5, and Hebrews 1:8). King David and then Jesus, who is the ultimate fulfillment of this verse, were both in the genealogical line of Judah. The phrase "gathering of the people" is best translated as "the obedience of the people" (the Jewish Masoretic Text-based "Holy Scriptures" renders it "obedience," as do the NAS, NIV, and NKJ translations). Although at His first coming Jesus' "kingdom" was not of this world (John 18:33-37), this verse mainly looks to His kingship upon earth at His second coming and His glorious millennial reign as King Messiah.

During Jesus' lifetime the authority to administer capitol punishment was taken away from Jewish rule by the Romans. For this reason the Jewish religious leaders appealed to Pilate to sentence Jesus to death, since they no longer had the authority. This authority is embodied in the word "sceptre" (a symbol of the right to rule) in Genesis 49:10.
